A Usefulness of BACTEC 460 on the Identification and Drug Susceptibility Test on Mycobacterium

Abstract
Early isolation of mycobacteria and differentiation of M. tuberculosis complex from other mycobacteria are very important in the diagnosis, treatment and assessment of prognosis of tuberculosis. But their completion requires long time. Therefore rapid radiometric method for both recovery of mycobacteria from chnical specimens and drug susceptibility testing have been developed. This study was designed to compare conventional method with BACTEC method with 30 species of M. tuberculosis and 6 different mycobacteria other than M. tuberculosis in order to evaluate usefulness of BACTEC method m chmcal application. The results are summerized as follows : 1. The time required for the primary isolation of M. tuberculosis was 10 to 20 days(mean 14. 2 days) and 3 to 12 days (mean 5.0 days) in conventional and BACTEC method, respectively. . The time required for the primary isolation of MOTT was 5 to 16 days(mean 11.3 days) and 3 to 5 days(mean 4.2 days) in conventional and BACTEC method, respectively. 3. The time used for the differentiation of M. tuberculosis from other mycobacteria with NAP test was 1 to 3 days. 4. The time of drug susceptibility test was 20 to 30 days(mean 24.5 days) and 5 to 10 days (mean 5.4 days) in conventional and BACTEC methods respectively. 5. The rate of susceptibility in each drug was significantly different. in both methods: Isoniazid shows 22.5 % and 79% ; ethambutol, 47.5% and 86.7% ; rifampin, 57.5% and 83.3% ; streptomycin, 72.5% and 83.3% in conventional and BACTEC methods, respectively.
